This topic is actually about two things: one: what i believe from a professional writer and deadpool fans perspective on what the plot will be and two: to tell you all that the Deadpool spin-off is not gonna follow the Wolverine movie and is a reboot so that means no Weapon XI bullshit.
possible plot: Wade Wilson loves what he does, he gets to travel the world, meet people then kill them, and getting paid for it too! But Wade comes down with cancer. After seeing him in action the Weapon X program kidnaps him, and for months tortures him but also turning him into a more dangerous person, recieving a mutant healing factor that cures his cancer and the ability to teleport, Wade is an unstoppable killing machine working side by side with black tom cassidy and others. But the scientists continue to torture him until he makes his way onto the other inmates "Dead Pool" list which is the inmates form of entertainment by betting on when someone will die however the tests run on Wade scar his face but his healing factor cannot heal the scars and Wade goes insane taking on the name of Deadpool and escaping his captors and eventually becomes friends with the hacker known as Weasel. Years ago Wade was hired to kill a blind woman named Althea or Blind Al but instead killed everyone where she was stationed except her, after going insane due to the Weapon X program Deadpool meets Al again and kidnaps her. Now living with both Weasel and Blind Al, Deadpool resumes the life of a mercenary, due to his insanity Deadpool is never serious always an annoying jerk. Furious over his escape the men in charge of Weapon X hire mercenaries among them Black Tom Cassidy, T-Ray, and the Black Swan to find and kill Deadpool. Deadpool does not see the big picture and decides to kill them all in an act of revenge for attacking his friends and home and out of boredom and to maintain his spot as the best in the killer for hire buisness Deadpool goes on a rampage towards Weapon X, taking out anyone in his way and making jokes while he does it.
Thats pretty much the plot i would use if i was writing it and thats probably the plot they'll use
